Studies on Genetic Monitoring through the Test of Biochemical Marker in Inbred Mice in Korea



Abstract
These studies were carried out to establish the genetic monitoring system and to survey genetic contamination of inbred mice which are being maintatined in Korea by checking of biochemical markers with Titan III(Helena Labs.) electrophoresis.
It was tested 7 loci(Idh-1, Pep-3, Car-2, Hbb, Gpi-1, Es-1, Es-3) of biochemical markers, mainly critical subset(Idh-1, Car-2, Hbb, Gpi-1). Animals were purchased from two institutes and one commercial breeder.
In A facility, there were b and ab type in Pep-3 (Peptidase-3) of BALB/c, and ab type in Gpi-1 (Glucose phosphate isomerase-1) of C57BL/6. It seems that the colonies of BALB/c and C57BL/6 were genetically contaminated. In B and C facilities were homozygous as long as the loci which were tested in this study.
These results suggest that it is required to check on genetic contamination regularly for maintaining pure lines and for improving animal quality. Various genetic monitoring methods should be established to improve the repeatation, confidence and accuracy of experiment with animals.
